DSLR Photomicrograph Tips

1. Focusing
To install micorscope adapterm, camera lens need to be removed from camera body, and therefore
it is unable to adjust focus with camera lens. For focusing, please use focusing knobs of microscope.
Secondly, focal point for LCD monitor and microscope eyepiece do not match normally. Therefore,
try to adjust focusing with an image rendered in LCD monitor, not with an image you see in an eyepiece.
For this reason, DSLR with live-view feature is recommended. Additionally, connecting camera to
TV/PC monitor enable you to easily adjust focus with help of large screen.
2. Stabilizer
When capturing an image through a microscope, the slightest movement will cause the image to
get blur. To avoid a blurry image, please use a remote switch or a self-timer. Also if your camera
has vibration mode, set it on (for about 1 second).
3. Exposure Adjustment
It is easier to shoot with Aperture Priority Mode (dial A).
If the image is too dark or too bright adjust your Aperture Settings (±).
The higher apertures correspond to darker images. Lower apertures correspond to brighter images.
4. White Balance
When the captured image appears too bluish or reddish, please manually set the white balance.
First, adjust the lighting to desired level and fix. Second, remove any samples from the microscope stage.
Third, expose camera sensor to blank and white target to have camera recognizes it as white.
 * Use a glass (with no sample) as a target for biological microscope.
 * Use a white/gray plate as a target for stereo/metallurgical microscope.
Once white balance is manually adjusted, you will be able to capture the sample with accurate color tone.
Many DSLR model have Color Space feature to reproduce desirable color tone afterwords as well.
5. ISO Sensitivity
When you shoot at high ISO speeds, noise (such as dots of lights and banding) may become noticeable.
When captured image is too dark, use a slower shutter speed or change the microscope light volume.
This will enables to capture images with a low ISO speed.
6. Other
For biological microscopes, please set the condenser to appropriate position so that aperture is in
accordance with designated magnification. (* Please refer to your microscope manual for detailed instruction)
